| CD28 是 T 細(xì)胞表面的核心共刺激受體，屬于免疫球蛋白超家族（IgSF），其通過與 抗原呈遞細(xì)胞（APC）上的配體 CD80（B7-1）和 CD86（B7-2）結(jié)合，提供 T 細(xì)胞激活的 “第二信號(hào)”（共刺激信號(hào)），是適應(yīng)性免疫應(yīng)答的關(guān)鍵調(diào)控節(jié)點(diǎn)。CD28 信號(hào)缺失會(huì)導(dǎo)致 T 細(xì)胞失能（Anergy），而過度激活則與自身免疫病及移植物抗宿主?。℅VHD）相關(guān)。在 腫瘤免疫治療中，CD28 共刺激信號(hào)的恢復(fù)可增強(qiáng) CAR-T 細(xì)胞或 PD-1 抑制劑療效，成為 免疫調(diào)控的重要靶點(diǎn)。 CD28 激活對(duì) T 細(xì)胞的共刺激會(huì)啟動(dòng)信號(hào)級(jí)聯(lián)反應(yīng)，導(dǎo)致 AP-1 和 NFκB 轉(zhuǎn)錄因子激活 和核轉(zhuǎn)位。這些途徑顯著增強(qiáng) T 細(xì)胞細(xì)胞因子的產(chǎn)生，特別是 IL-2，從而促進(jìn) T 細(xì)胞增殖、 分化和存活。 | |
